Genetic characterization of Aberdeen Angus cattle using molecular markers Genet. Mol. Biol.
Vasconcellos,Luciana Pimentel de Mello Klocker; Tambasco-Talhari,Daniella; Pereira,Andréa Pozzi; Coutinho,Luiz Lehmann; Regitano,Luciana Correia de Almeida.
Aberdeen Angus beef cattle from the Brazilian herd were studied genetically using restriction fragment length polymorphism (RFLP) of the kappa-casein - HinfI (CSN3 - HinfI), beta-lactoglobulin - HaeIII (LGB - HaeIII) and growth hormone AluI (GH- AluI) genes, as well as four microsatellites (TEXAN15, CSFM50, BM1224 and BM7160). The RFLP genotypes were determined using the polymerase chain reaction (PCR) followed by digestion with restriction endonucleases and electrophoresis in agarose gels. With the exception of the microsatellite BM7160, which was analyzed in an automatic sequencer, the PCR products were genotyped by silver staining. The allele and genotype frequencies, heterozygosities and gene diversity were estimated. The values for these parameters of...

Genetic divergence among and within Colletotrichum lindemuthianum races assessed by RAPD Trop. Plant Pathol.
Talamini,Viviane; Souza,Elaine A.; Pozza,Edson A.; Silva,Gilvan F.; Ishikawa,Francine H.; Camargo Júnior,Osnil A..
Genetic divergence within and among races of Colletotrichum lindemuthianum was determined using RAPD markers. In addition to the different races of the fungus three isolates of the sexual stage of Colletotrichum lindemuthianum (Glomerella cingulata f.sp. phaseoli) were included in this study. The band patterns generated using 11 primers produced 133 polymorphic bands. The polymorphic bands were used to determine genetic divergence among and within the pathogen races. The isolates analyzed were divided into six groups with 0.75 relative similarity. Group VI, formed by three isolates of the sexual phase of Colletotrichum lindemuthianum, was the most divergent. Races previously determined using differential cultivars did not correlate with the results...

Genetic diversity of four cattle breeds using microsatellite markers R. Bras. Zootec.
Machado,Marco Antonio; Schuster,Ivan; Martinez,Mário Luiz; Campos,Ana Lúcia.
Four cattle breeds (Gyr, Nellore, Guzerat and Holstein) were analyzed by amplification of genomic DNA using microsatellite loci to evaluate the genetic diversity within and among them. DNA samples of 18 animals from each breed were collected to access the genetic content of them. Allele frequencies were calculated and used to generate a Nei's genetic distance matrix what was used to build a dendrogram following UPGMA clustering. As expected, Holstein breed was the most distinct from the other breeds: 1.15 in relation to Gyr, 1.12 in relation to Nellore and 0.94 in relation to Guzerat. The closest genetic distance was 0.25 between Guzerat and Nellore. A total of 64 alleles in all four breeds were detected using nine microsatellite primers. Each breed showed...

Genetic improvement of beef cattle through opportunities in genomics R. Bras. Zootec.
Miller,Stephen.
Genomics will improve the efficiency of beef cattle genetic improvement programs through the incorporation of genomic predictions into traditional genetic evaluations. The global dairy cattle breeding industry has been changed considerably in the last year through the implementation of genomic selection. Now proven to work in dairy cattle breeding, the challenge remains for the beef industry to successfully implement this technology. The primary challenge in beef cattle is the required resource population that relates genomic profile to phenotypic performance, which is quite large and its establishment will require collaboration or a significant investment by any one enterprise. Another challenge in beef cattle is the requirement for genomic predictions to...

Hidden Markov models applied to a subsequence of the Xylella fastidiosa genome Genet. Mol. Biol.
Silva,Cibele Q. da.
Dependencies in DNA sequences are frequently modeled using Markov models. However, Markov chains cannot account for heterogeneity that may be present in different regions of the same DNA sequence. Hidden Markov models are more realistic than Markov models since they allow for the identification of heterogeneous regions of a DNA sequence. In this study we present an application of hidden Markov models to a subsequence of the Xylella fastidiosa DNA data. We found that a three-state model provides a good description for the data considered.

Identificação de Brucella spp. em bovinos com lesões sugestivas de brucelose. Infoteca-e
ROSINHA, G. M. S.; SANTOS, L. R. dos; ELISEI, I.; SANCHES, C. C.; BERTOLACCI, M. A. B. C.; MANTOVANI, C.; ELISEI, C.; SOARES, C. O..

Identification of Pm24, Pm35 and Pm37 in thirteen Egyptian bread wheat cultivars using SSR markers Ciência e Agrotecnologia
Emara,Hassan Mahmoud; Omar,Ayman Feisal; El-Shamy,Moustafa Mahmoud; Mohamed,Mona Elsaid.
ABSTRACT Powdery mildew of wheat (Triticum spp.) caused by Blumeria graminis f.sp. tritici (DC) E.O. Speer Em. Marchal is one of the most important bread wheat diseases in Egypt. All the Egyptian common bread wheat cultivars are susceptible to that disease at seedling and adult stages. Breeding of resistant cultivars is the most economical and environmentally safe method to eliminate the disease and reduce crop losses. Combinations of two or more effective resistance genes may lead to better, more durable resistance to that disease. Eight Pm genes i.e. Pm2, Pm6, Pm12, Pm16, Pm24, Pm35, Pm36 and Pm37 out of 21 powdery mildew monogenic wheat lines (Pm) were resistant to 42 individual isolates of powdery mildew collected from different governorates in the...

Identification of skipjack tuna juveniles based on DNA control region sequences and potential spawning area around reunion island ArchiMer
Nikolic, Natacha; Jerome, Marc; Fonteneau, Alain; Evano, Hugues; Verrez-bagnis, Veronique.
In this study, we confirmed that DNA control region can be used to identify potential spawning area from pelagic juveniles species. We used the mitochondrial DNA control region to identify tuna juveniles predated by common dolphinfish and sampled in surface waters around Reunion Island. These were skipjack juveniles at an early stage (small length and young age). The biological results confirmed the species identification based on DNA analysis. Catch data by longline reveal a higher number of skipjack from October to December around Reunion Island corresponding to the period of our skipjack juvenile sampling

In situ QCM DNA-biosensor probe modification ArchiMer
Lazerges M, M; Perrot, H; Zeghib N, N; Antoine, Elisabeth; Compere, Chantal.
We report on a probe modification of a quartz crystal microbalance (QCM) DNA-biosensor that permits to reversibly change the DNA sequence detected. A QCM DNA-biosensor was designed by immobilization of a 20-base DNA-disulfide probe on the gold-covered quartz surface of a 27 MHz microbalance (9 MHz, third overtone). After immobilization on the gold covered quartz surface, this probe was modified by hybridization with a 45-base DNA that includes the complementary 20-base sequence, a 5-base spacer and a non-complementary 20-base sequence. The non-complementary sequence constitutes a new probe, different from the DNA-disulfide probe, that permits the detection of a new DNA target. As this 45 bases DNA is changeable by dehybridization, successive different DNA...
